Change in volumetric tumor growth rate after cytotoxic therapy is predictive of overall survival in recurrent glioblastoma

چکیده
Abstract Background Alterations in tumor growth rate (TGR) recurrent glioblastoma (rGBM) after treatment may be useful for identifying therapeutic activity. The aim of this study was to assess the impact volumetric TGR alterations on overall survival (OS) rGBM treated with chemotherapy or without radiation therapy (RT). Methods Sixty-one patients concomitant (RT) at 1 st 2 nd recurrence were retrospectively examined. Pre- and post-treatment contrast enhancing volumes computed. Patients considered “responders” if they reached progression-free 6 months (PFS6) showed a decrease “non-responders” didn’t reach PFS6 increased. Results Stratification by based resulted significant differences OS both all RT (P < 0.05). A = 0.002), smaller baseline volume 0.008), O 6-methylguanine-DNA methyltransferase promoter methylation 0.03) fewer number recurrences 0.04) significantly associated longer controlling age, sex RT. Conclusion PFS6, along volume, radiation. Importantly, that exhibited also measurable TGR.

Increase in tumor-associated macrophages after antiangiogenic therapy is associated with poor survival among patients with recurrent glioblastoma.
Antiangiogenic therapy is associated with increased radiographic responses in glioblastomas, but tumors invariably recur. Because tumor-associated macrophages have been shown to mediate escape from antiangiogenic therapy in preclinical models, we examined the role of macrophages in patients with recurrent glioblastoma. We compared autopsy brain specimens from 20 patients with recurrent glioblas...
متن کاملOverall survival benefit from surgical resection in treatment of recurrent glioblastoma.
ESMO Clinical Practice Guidelines on high-grade gliomas dismiss impact of surgical resection on overall survival (OS) in treatment of recurrent glioblastoma (GBM), citing two pooled analyses as evidence of no effect. Neither analysis considered one critical factor in analyzing efficacy of re-resection—the extent of resection (EOR)—known to be important at first surgery. Furthermore, only 12% (3...
متن کاملProscillaridin A is cytotoxic for glioblastoma cell lines and controls tumor xenograft growth in vivo
Glioblastoma is the most frequent primary brain tumor in adults. Because of molecular and cellular heterogeneity, high proliferation rate and significant invasive ability, prognosis of patients is poor. Recent therapeutic advances increased median overall survival but tumor recurrence remains inevitable. In this context, we used a high throughput screening approach to bring out novel compounds ...

Survival and functional status after resection of recurrent glioblastoma multiforme.
OBJECTIVE To determine the selection factors for and results of second resections performed to treat recurrent glioblastoma multiforme (GM), we studied 301 patients with GM who were treated from the time of diagnosis using two prospective clinical protocols. METHODS The patients were prospectively followed from the time of diagnosis, using clinical and radiographic criteria after maximal surg...
